A study published in July 2025 showed that prescribing gabapentin to patients with chronic lower back pain is associated with an increased risk of dementia and cognitive impairment, especially when receiving six or more prescriptions of the drug. A parallel study from September 2025 on aged mice confirmed the negative effect of long-term gabapentin intake on cognitive functions through the mechanism of tau protein hyperphosphorylation and disruption of the Sirt1-CaMKIIa signaling pathway in the hippocampus. On the other hand, a multicenter retrospective study (May 2025) revealed an unexpected positive effect: patients with glioblastoma who took gabapentin after surgical tumor resection showed an increase in overall survival by 4-6 months compared with the control group, which is associated with inhibition of thrombospondin-1 (TSP-1). The study also showed that gabapentin reduces serum TSP-1 levels, which may serve as a potential biomarker of a therapeutic response. [source]
